Properties of particles in sap
TIP:
40-50.16-60 °C. LIV: 0.01-14.27-72 days. DEP: log10 minus 1 (3/6), or 2-3
(2/6), or 4 (1/6). Infectivity of sap not changed by treatment with di-ethyl
ether (2/2). Leaf sap contains few virions (19/20), or contains many virions
(1/20).
Particle morphology
Virions geminate (33/33), or
unusually shaped (1/33); not enveloped (19/19); 15-19.12-25 nm in diameter;
15-30.41-52 nm in length; rounded in profile (5/9), or angular in profile (5/9);
with a conspicuous capsomere arrangement (1/12), or without a conspicuous
capsomere arrangement (11/12).
Physical properties
One sedimenting component in purified
preparations (6/9), or two sedimenting components in purified preparations
(3/9), or three sedimenting components in purified preparations (1/9);
sedimentation coefficient 69-78.6-90 S; of the other(s) 50-56.66-70
S. Density 1.717 g cm-3 in CsCl. Isoelectric point pH 7.
A260/A280 ratio 1.36-1.376-1.4.
Biochemical properties
Virions contain 18-20-22 % nucleic
acid; 78-80-82 % protein; 0 % lipid.
Genome consists of DNA (23/23); single-stranded (22/22); circular
(16/16). Total genome size 5.096-5.286-5.57 kb. Genome of two parts (15/15);
largest (or only) genome part 2.588-2.678-2.87 kb; the 2nd largest
2.508-2.627-2.724 kb. Base composition 21-22.31-23.5 % G; 26-27.76-29 % A;
18.3-18.71-19.8 % C; 30.2-31.2-32.6 % T. Infectivity retained when deproteinised
with proteases (2/4), or decreased when deproteinised with proteases (2/4);
retained when deproteinised with phenol or detergent (5/5). Poly A region absent
(3/3). Additional factor not required for infectivity (4/4). Genome has no
tRNA-like activity (2/2).
Features of the genome
Non-genomic nucleic acid
found in the virions (2/6), or not found in the virions (4/6); is of an unusual
type of structure (1/1). Sub-genomic mRNA found in infected cells
(1/1).

Virion protein(s) one (6/7), or
two (1/7); Mr 27400-29950-34000. Mr of 2nd largest 27000.
Virion proteins not glycosylated (1/1).
Virus-coded non-virion proteins identified by genomic sequence
analysis (2/2); five proteins found (2/2). Mr 40100-40200-40300.
Mr of 2nd largest 32700-33200-33700. Mr of 3rd
29300-29450-29600. Mr of 4th 15600-15700-15800. Mr of 5th
and smaller 14500-14800-15100.
Replication
Genome replicates in nuclei (6/6).
Replication does not depend on a helper virus (8/8).
Cytopathology
Virions found in cytoplasm (5/16), or in
nuclei (15/16), or in nucleoli (2/16), or in chloroplasts (1/16), or in cell
vacuoles (1/16). Inclusions present in infected cells (12/13), or absent from
infected cells (1/13); are crystals in the nucleus (3/12), or amorphous
X-bodies (2/12), or viroplasms (1/12), or unusual in shape (11/12); they
contain virions (5/6), or they do not contain virions (2/6).
